He was born in the Jabal al-Mukaber neighborhood of Jerusalem in 1950.
Early in his academic career, he studied under Sheikh Hashem al-Baghdadi, one of the scholars of the Qur’anic recitations in Jerusalem, learning from him the rules of recitation and intonation.
He graduated from Al-Aqsa Sharia High School in 1969, and obtained a BA from the Faculty of Sharia at the University of Jordan in 1973, followed by a MA in Contemporary Islamic Studies from Al-Quds University.
He has participated in numerous conferences in Arab, Islamic, Asian and American countries, and has appeared in dozens of television programs and interviews.
After graduating from the Faculty of Sharia in 1973, he worked as a teacher at the Dar al-Aytam Islamic School, then moved to teach at Al-Aqsa Sharia High School, then became the principal of the Sharia school, then a supervisor of Islamic guidance at the Islamic Endowments Department in Jerusalem.
He began teaching and preaching at Al-Aqsa Mosque in 1982, then took over the management of Al-Aqsa affairs from 1986 to 2006.
Between 1987 and 1990, he was appointed a member of the Islamic Endowments Council in the West Bank, in addition to heading the Supreme Fatwa Council in Palestine. He is a founding member of several charitable organizations.
He served as Chairman of the Advisory Board of the College of the Qur'an and Islamic Studies, a founding member of the Council of Scholars and Preachers in Jerusalem, a member of the International Islamic Fiqh Academy in Saudi Arabia, and the Board of Trustees of the Martyr Yasser Arafat Foundation.
He served as General Supervisor of Al-Israa Magazine, published by the Directorate of Information and Islamic Research at the Palestinian Fatwa House. Palestinian President Mahmoud Abbas appointed him Mufti of Jerusalem and the Palestinian territories with the rank of minister in 2006.
Following Israel's adoption of the force-feeding law for prisoners, the Sheikh issued a fatwa permitting prisoners' resistance to force-feeding, stating that "its victims are martyrs," adding that this method is permissible "if the prisoners find no other means to achieve their legitimate human rights."
Throughout his professional and academic career, Sheikh Muhammad Hussein received several medals and awards, including a medal from President Hosni Mubarak in 1990, a medal from the late President Yasser Arafat in 1997, and the Palestine Prize in Arts and Humanities in 2000.
